This chapter presents an introduction to the kinetic analysis of SPR biosensor data for the determination of affinity and kinetic rate constants of biomolecular interactions between an immobilized and a soluble binding partner. For binding to occur in SPR assays the analyte must be transported laterally (diffusion) in the flow cell from the bulk solution to the surface before any interaction with a ligand can take place (Figure 3). The figure below shows the relation between the kinetics and the response, time to equilibrium and the analyte concentration.
